The gastrointestinal (GI) tract is a physiological and chemical barrier and thus poses complex demands on oral therapy. The fact that about 50% of newly developed active ingredients are either poorly soluble or completely insoluble in water, leads to additional challenges because of their poor absorption rates and low bioavailability. Another problem is the low absorption rate of some active ingredients. It is extremely difficult to develop potent formulations with such active ingredients. One frequent approach is the use of dissolving techniques, excipients and lipids. Phospholipid-based formulations can positively impact the absorption of active ingredients through the following mechanisms:
- modification of drug release
- increase of bioavailability
